NEW & NOTEWORTHY Endothelin-1 (ET-1) is recognized as the body’s most potent endogenous vasoconstrictor, but the impact of this peptide on vascular function is not well understood. The present study revealed that the intra-arterial administration of ET-1 impaired both microvascular and conduit vessel function of the leg in young, healthy, humans.

Endothelin-1 (ET-1), the most potent vasoconstrictor known, was first described in 1988. 1 The endothelin family also includes endothelin-2, endothelin-3, and the sarafotoxins. The endothelins are widely distributed in tissues, produced by endothelial cells and many epithelial cell types. 2-5 Under normal conditions, ET-1 apparently maintains vascular tone and exerts its action in a variety of Endothelin-1 critically influences cardiac function via superoxide-MMP9 cascade Catherine K. All three endothelins are classified as having vascular homeostasis functions.

Endothelins are peptides with receptors and effects in many body organs. Endothelin constricts blood vessels and raises blood pressure. The endothelins are normally kept in balance by other mechanisms, but when overexpressed, they contribute to high blood pressure, heart disease, and potentially other diseases. Endothelins are 21-amino acid vasoconstricting peptides produced primarily in the endothelium having a key role in vascular homeostasis. Endothelins are implicated in vascular diseases of
